Natural immunity boosters: Clinical trial reports
Background
Balanced nutrition containing the right proportion of macronutrients and micronutrients has been providing strong and robust immunity to humans since evolution. But the modern and fast going lifestyle has resulted in malnutrition and attenuated the immunity of Humans in general; as a result, various infections caused by fungi, bacteria, viruses and other pathogens are becoming life-threatening and increasing the burden on the health care system as is evident from the current COVID-19 pandemic. Hence, to cope up with the alarming situation, proper and balanced nutrition and become essential, which is in the current scenario available in most of the food supplements and Heath drinks. The Health drinks contain several vitamins, minerals and extracts from natural plants and herbs which have immune-boosting, antiviral, antioxidant, anti-inflammatory effects. These include Vitamin A, B, C, D, E, K, folic acid, zinc, selenium, magnesium, calcium, curcumin, probiotics, lactoferrin, quercetin, etc. One such health drink is the Mulmina® Mango which contains vitamins, minerals and mesonutrients such as extracts from mango, Centella Asiatica and turmeric. Mango extracts are rich in Mangiferin which has different pharmacological properties like antioxidant, anti-inflammatory, antimicrobial, antidiabetic, antiallergic, anticancer, hypocholesterolemic, and immunomodulatory. Also, the vitamin C present has high antioxidant property that reduces oxidative stress and prevents cell damage. Hence, in-vitro clinical trials were carried out to prove the efficacy of the health drink1.

Method 1:
The Primary dendritic cells were exposed to the toxic substance LPS followed by treatment with Mulmina® Mango for 4hours and the cell supernatant collected, The cell-supernatant bound immune markers such as TNF-α, IL-1β and MIP-1 were then estimated.
Results 1
Mulmina® Mango showed inhibition of LPS-induced TNF-α by 54.46±1.3%, LPS-induced IL-1β generation by 55.60±0.5% and also MIP-1-α generation by 44.04±3.3 at a dose of 800 μg/mL,
Mulmina® Mango exhibited increased phagocytic potential of RAW macrophages by 54.65±3.1% at 800μg/mL over the control.
Discussion
Hyper-inflammatory response induced by SARS-CoV-2 is a major cause of the complexities arising in COVID-19 patients.2 It is well known that some of the pro-inflammatory cytokines like IL-1β, IL-6, and TNF-α and chemokines which are up regulated including MIP-1α, MCP-1 and GRO/KC are involved in the process of pathological pain. Results from different studies have shown that COVID-19 infected patients have shown increased levels of IL-2, IL-7, G-CSF, CXCL10, MCP-1, MIP-1α, TNF-α, and IL-6, which result in severe hyper inflammation-causing lung damage, ARDS and more related complications. Hence, determining the levels of these immune biomarkers become important for evaluating the effectiveness of any treatment 3,4. In the current study, Mulmina® Mango showed inhibition of LPS-induced TNF-α, IL-1β generation and also MIP-1-α generation thus proving the effectiveness in protecting the dendrocytes1.
Studies reveal that the generation of phagolysosomes occurs in the process of phagocytosis and it creates an unfavorable environment for most microorganisms, thus helping in preventing infection. Phagocytes such as macrophages form reactive nitrogen species (RNS) which lead to the destruction of microbes, pathogens and limits the spread of infection 5. Thus, the significant increase in the phagocytic potential of macrophages as compared to control in a dose-dependent manner reflects the immunomodulatory effect of Mulmina® Mango 1.
Conclusion
The above studies show that Mulmina® Mango exhibits good antioxidant, anti-inflammatory properties and immunomodulatory effects which can prove to be effective in boosting immunity, reducing the COVID-19 related complexities and also help to reduce the burden on the health care system.
